As reported in Nature Communications, Cellular & Molecular Immunology, Cell Reports, and Allergy our 2-in-1 microarray contains both full-length proteins and VirScan™ peptides spanning the S1 and S2 spike of SARS-CoV-2. This allows analysis of both antibodies binding full-length proteins and determining our VirScan™ epitope-level granularity across the important ACE2 receptor binding spike.  To ensure solubility, the peptide scan utilizes BSA-conjugated overlapping 12-mer peptides covering the entire S1 and S2 spike protein of SARS-CoV-2, the virus that causes pandemic COVID-19 disease. These deep immune profiling details are useful for coronavirus vaccine development, developing therapeutic antibodies, profiling convalescent plasma, and more.

Format: 14 sub-arrays (2X7 format). Each sub-array contains the following full length SARS-CoV-2 proteins (E-protein, N-protein, NSP1, NSP2, NSP4, NSP5, NSP7, NSP8, NSP9, NSP10, RdR, NSP14, NSP15, S1, RBD, S2 ) and 12-mer VirScan™ peptides (conjugated to BSA) covering the S1 and S2 proteins.

Manufacture: Non-contact piezoelectric inkjet on coated glass (PATH®); fluorescence detection.

Protein Expression System: E. coli.

Controls: Cy3, Cy5, BSA, eGFP, human IgG, human IgM, buffer only.

Storage: -80˚C in bags with plastic slide holders and desiccant.

2-in-1 microarray service details & data deliverables.

A typical 2-in-1 microarray service involves case and control serum samples diluted 1:1000 and reacted with individual microarrays with blocking buffer. After incubation, samples are stained for one or two desired fluorescent secondary detection antibodies (ie IgG / IgA / IgM). Arrays are scanned and raw TIF images are created using Molecular Devices Genepix microarray scanners. Spots are then aligned to CDI library (.gal) files, and spot readings are output as raw Genepix results (.gpr) files. These raw results are then delivered both as .gpr files and combined data tables.

2-in-1 microarray service sample requirements.

Human serum or plasma: Send one 20μL aliquot per sample.

Antibody isolates: At least 3μg per sample (conc. >0.1mg / mL).

Human (CSF) cerebrospinal fluid: At least 1.5mL per sample.

Protein / peptide / small molecule: At least 3μg per sample.

Sample shipping & sample return.

We will include shipping details in your quote – you must cover the cost of shipping samples to CDI. Typically, after you receive your report, CDI keeps the remaining samples for two months and then disposes of them. When shipping to us, please let us know if you want the remaining samples returned after the study is complete. We will charge for return shipping.
